APLF (Aprataxin and PNK-like factor) nuclease is involved in the repair of single-stranded and double-stranded DNA breaks. The DNA damage site was recruited by interacting with poly (ADP- ribose). The enzyme is a kind of post-translational modification synthesized briefly at the chromosome damage site, which can accelerate the repair of DNA strand breaks. The activities of purine-pan-pyrimidine nucleoside (AP) endonuclease and 3'-5' exonuclease were shown in vitro. Incisions can also be introduced when hydroxyuracil and other types of pyrimidine bases are destroyed. Together with PARP3, it can promote the chromatin retention of LIG4-XRCC4 complex and accelerate the DNA connection during the nonhomologous end joining (NHEJ).
