Bruton tyrosine kinase (BTK), is a 77 kDa protein containing 659 amino acids, which can be divided into five different functional domains: Pleckstrin homologous (PH) domain is located near the N-terminal, followed by Tec homologous (TH), Src homology 3 (SH3), SH2 and SH1 or tyrosine kinase (TK) domain. BTK was initially found to be a key mediator of B cell receptor signals during acquired immune development and function. More and more evidence shows that BTK plays multiple roles in monocytes of the innate immune system, especially in dendritic cells and macrophages, and plays an important role in the proliferation and survival of leukemic cells in many B-cell malignant tumors.
