NEK3 (NIMA Related Kinase 3) belong to a large family of serine/threonine (Ser/Thr) kinases, which play an important role in coordinating microtubule dynamics during cilia development and mitosis. Diseases associated with NEK3 include muscle phosphorylase kinase deficiency and glycogen storage disease Ia. NEK3 regulates microtubule acetylation and promotes prolactin-mediated phosphorylation of PXN and VAV2 by affecting protein kinases that affect the morphology and polarity of neurons. It participates in the cytoskeleton recombination and movement of breast cancer cells mediated by prolactin by involving the mechanisms of RAC1 activation and PXN and VAV2 phosphorylation.
