Introduction
In the past 30 years, cryogenic electron microscopy (Cryo-EM) has revealed the fine structures of ribosomes, membrane proteins and other key cellular proteins. Structural biologists say it is no exaggeration to say that cryogenic electron microscopy is in the midst of a revolution: cryogenic electron microscopes can quickly generate high-resolution molecular models far beyond methods such as X-ray crystal diffraction. Protein structure is firmly regarded as a fundamental cornerstone of drug discovery platforms.
Application
Cryo-EM has emerged as a promising new technique to obtain structural data from challenging proteins. Recently Bulatov et al. performed an exhaustive search within the PDB for crystal structures related to E3 ligases that can be very useful for drug discovery perspectives. The latest developments in detector technology and software algorithms have allowed cryogenic electron microscopes to determine biomolecule structures close to atomic resolution. As an alternative to X-ray crystallography or NMR spectroscopy, it has aroused widespread concern that macromolecular structures can be determined without crystallization, which plays an important role in PROTAC development.
