SMCC-Plus
SMCC-Plus is a bifunctional, thiol-reactive heterobifunctional linker commonly used in targeted protein degradation workflows to connect a maleimide-containing module to a thiol-bearing partner. Structurally, it provides a maleimide handle for rapid and selective conjugation with cysteine thiols, while the complementary reactive group enables formation of a stable linkage to the other PROTAC component, yielding a defined spacer that preserves bioactive conformations. In PROTAC design, such linkers are critical because they control the effective distance and relative orientation between the ligand that recruits the E3 ligase and the ligand that binds the target protein, thereby tuning ternary complex formation and degradation potency. SMCC-Plus is therefore valuable for constructing modular degraders, performing site-specific bioconjugation, and enabling systematic linker optimization in experimental studies focused on targeted degradation mechanisms and structure–activity relationships.

SMCC-Plus is a bifunctional PROTAC linker designed to connect a ligand-bearing warhead to an E3 ligase–binding module through robust, chemoselective coupling. Its functional-group architecture supports reliable assembly of targeted protein degraders, enabling modular synthesis and optimization of linker length and attachment points. The following points describe the linker’s structure and practical reactivity considerations in detail below.
Structure: SMCC-Plus is characterized by a maleimide-containing electrophilic handle paired with a complementary amine-reactive functionality. It incorporates conjugated unsaturation suitable for stable bioconjugation and contains heteroatom-rich motifs that support aqueous compatibility and controlled reactivity. The linker presents a defined, modular scaffold with functional groups positioned for PROTAC assembly.
Reactivity: The linker is typically used in stepwise PROTAC construction via chemoselective coupling of the maleimide electrophile to thiol-containing partners, followed by amine-directed conjugation where applicable. Reactions are commonly performed under mild, buffered conditions to preserve protein-ligand integrity, using standard organic solvents for solubility. Thiol–maleimide addition proceeds through nucleophilic attack and rapid Michael-type addition, while amine coupling relies on electrophile activation and nucleophilic substitution.
